目的 探讨SCID鼠与免疫学相关的外周血指标变化及个体“渗漏”问题。方法 采用血细胞全自动分析仪检测SCID、NC、BALB/c鼠 14项外周血指标 ,3H TdR掺入测T、B淋巴细胞转化功能和细胞毒活性。结果 SCID鼠与NC、BALB/c鼠相比 ,WBC、淋巴细胞分类 (Ly)的比例显著减少 ;中性粒细胞 (GR)和红细胞体积分布宽度 (RDW )显著增加。随着鼠龄的增长 ,WBC和Ly增加 ,GR和RDW减少 ,其中变化最显著的个体 (WBC≥ x±2SD)称之为“渗漏” ,在鼠龄 6~ 8周时和 17~ 19周时的渗漏率分别为 5 .5 %和 2 6 .7%。此外 ,SCID鼠的T、B淋巴细胞转化和CTL细胞功能与BALB/c鼠相比显著低下。结论 SCID鼠T、B淋巴细胞联合缺陷 ,可在免疫生物学领域中广泛应用 ,但要及时剔除个体“渗漏”鼠
Objective To investigate the changes of immune related peripheral blood markers and individual “leakage” in SCID mice. Methods 14 peripheral blood markers of SCID, NC and BALB / c mice were detected by automatic blood cell analyzer, T, B lymphocyte transformation and cytotoxicity were measured by 3H TdR incorporation. Results Compared with NC and BALB / c mice, the percentage of WBC and Lymphocytes was significantly decreased in SCID mice. The width of neutrophil (GR) and erythrocyte volume distribution (RDW) increased significantly. WBC and Ly increased with the increase of mice’s age, and GR and RDW decreased. Among them, the most significant change (WBC≥x ± 2SD) was called “leakage”. At the age of 6-8 weeks and 17-19 Week of leakage rates were 5.5% and 26.7%. In addition, T, B lymphocyte transformation and CTL cell function in SCID mice were significantly lower compared to BALB / c mice. Conclusions The combined defects of T and B lymphocytes in SCID mice are widely used in the field of immunobiology. However, it is necessary to eliminate the “leakage” mice
