该文探讨白头翁汤正丁醇提取物(butyl alcohol extract of Baitouweng decoction,BAEB)对白念珠菌细胞膜的抑制作用。以Spot assay观察BAEB对白念珠菌细胞活性的影响;酶标仪检测BAEB干预后白念珠菌细胞内渗透压变化;荧光显微镜观察BAEB对白念珠菌细胞膜通透性的影响;高效液相检测白念珠菌细胞膜麦角甾醇的含量;q RT-PCR法检测细胞膜麦角甾醇生物合成相关基因的表达。结果显示,256,512,1 024 mg·L~(-1)BAEB干预下白念珠菌活性逐渐降低;512,1 024 mg·L~(-1)BAEB组白念珠菌胞内甘油含量显著性增多(P<0.05),与白念珠菌胞内渗透压相关的基因HOG1分别下调9.1,9.3,5.5倍;512,1 024 mg·L~(-1)BAEB组白念珠菌红色荧光细胞明显增多;1 024 mg·L~(-1)BAEB干预后麦角甾醇峰面积为35.884 95,有显著性差异(P<0.05);1 024 mg·L~(-1)BAEB干预组ERG1,ERG2,ERG3,ERG4,ERG5,ERG6,ERG10,ERG11,ERG13,ERG24,ERG25,ERG251,ERG26与UPC2分别下调6.58,4.89,4.15,9.24,3.41,9.84,3.08,7.50,5.53,5.90,2.45,3.25,1.98,10.07倍。BAEB可通过抑制麦角甾醇及其生物合成相关基因的表达,进而抑制白念珠菌细胞膜完整性。
This article discusses the inhibitory effect of butyl alcohol extract of Baitouweng decoction (BAEB) on Candida albicans cell membrane. The effects of BAEB on the cell viability of Candida albicans were observed by Spot assay. The changes of cell osmolality of Candida albicans after BAEB intervention were detected by microplate reader. The effect of BAEB on the cell membrane permeability of Candida albicans was observed by fluorescence microscopy. Cell membrane ergosterol content; q RT-PCR method to detect cell membrane ergosterol biosynthesis related gene expression. The results showed that the activity of Candida albicans decreased with the doses of 256,512 and 1 024 mg · L -1 of BAEB. The intracellular glycerol content of Candida albicans group was significantly increased in the groups of 512 and 1 024 mg · L -1 BAEB P <0.05). The HOG1 gene, which was related to the intracellular osmotic pressure of Candida albicans, was down-regulated by 9.1, 9.3 and 5.5 times respectively. The number of Candida albicans red fluorescence cells in 512, 1 024 mg · L -1 BAEB group was significantly increased. There was a significant difference (P <0.05) in the ergosterol peak area after treatment with 024 mg · L -1 BAEB (P <0.05). The ERG1, ERG2, ERG3, ERG4 , ERG5, ERG6, ERG10, ERG11, ERG13, ERG24, ERG25, ERG251, ERG26 and UPC2 were down-regulated 6.58,4.89,4.15,9.24,3.41,9.84,3.08,7.50,5.53,5.90,2.45,3.25,1.98,10.07 times . BAEB can inhibit Candida albicans cell membrane integrity by inhibiting the expression of ergosterol and its biosynthesis-related genes.
