目的研究不同分子量的罗非鱼鱼鳞胶原蛋白多肽对体外培养新生小鼠颅骨MC3T3-E1细胞增殖与Ⅰ型胶原蛋白分泌及Ⅰ型胶原蛋白mRNA表达的影响。方法采用WST-8法测定胶原蛋白多肽对小鼠成骨细胞株MC3T3-E1细胞增殖的影响,ELISA法检测胶原蛋白多肽对MC3T3-E1细胞Ⅰ型胶原蛋白分泌的影响,采用real-time PCR分析不同分子量的胶原蛋白多肽对Ⅰ型胶原蛋白mRNA表达的影响。结果胶原蛋白多肽能促进MC3T3-E1细胞增殖(P<0.01)、促进MC3T3-E1细胞Ⅰ型胶原蛋白的分泌和Ⅰ型胶原蛋白mRNA的表达(P<0.05,P<0.01)。结论罗非鱼鱼鳞胶原蛋白多肽能有效促进MC3T3-E1细胞增殖与Ⅰ型胶原蛋白分泌,分子量较小的多肽(917.3Da)作用更加明显。
Objective To study the effects of different molecular weight Tilapia fish scale collagen peptides on the proliferation, the type I collagen secretion and the type I collagen mRNA expression of MC3T3-E1 cells cultured in vitro. Methods The effect of collagen polypeptide on the proliferation of mouse osteoblast cell line MC3T3-E1 was assayed by WST-8 method. The effect of collagen polypeptide on the type Ⅰ collagen secretion of MC3T3-E1 cell line was detected by ELISA. Real-time PCR analysis The Effect of Different Molecular Weight Collagen Peptides on the Expression of Type Ⅰ Collagen mRNA. Results Collagen peptide could promote the proliferation of MC3T3-E1 cells (P <0.01) and promote the secretion of type Ⅰ collagen and type Ⅰ collagen mRNA in MC3T3-E1 cells (P <0.05, P <0.01). Conclusion Tilapia fish scale collagen polypeptide can effectively promote the proliferation of MC3T3-E1 cells and the secretion of type Ⅰ collagen. The effect of polypeptide with smaller molecular weight (917.3Da) is more obvious.
