目的探讨骨桥蛋白(OPN)及高迁移率族蛋白B1(HMGB1)在子宫内膜异位症(EM)中的水平,观察载药输卵管节育器合化瘀止痛散对HMGB1的影响及对EM的治疗作用。方法 40只大鼠随机分为4组,每组各10只,实验组采用逆流法造模并放置可逆性输卵管节育器并于术后4周加用化瘀止痛散灌胃,对照组1采用逆流法造模后仅放置可逆性输卵管节育器(术后4周蒸馏水灌胃),对照组2逆流法造模术后4周用化瘀止痛散灌胃,空白对照组不做造模但以蒸馏水灌胃。采用放射免疫法分别检测各组大鼠造模前、造模后、治疗后血清中OPN及HMGB1水平的变化。结果与空白组(造模前)比较,各组大鼠造模后血清OPN、HMGB1的水平显著升高(P均<0.05);实验组和对照组不同程度的降低血清OPN和HMGB1的水平,与造模后比较差异均有统计学意义(P均<0.05);其中实验组和两个对照组比较其差异亦有统计学意义(P均<0.05)。结论 OPN、HMGB1在EM发生发展中起重要作用,载药输卵管节育器合化瘀止痛散可能通过调节OPN、HMGB1的水平以抑制异位内膜的增生,促进炎症包块的消退,并有很好的抗炎镇痛作用。
Objective To investigate the levels of osteopontin (OPN) and high mobility group box 1 (HMGB1) in endometriosis (EM) Therapeutic effect. Methods Forty rats were randomly divided into 4 groups (n = 10 in each group). The experimental group was established by countercurrent method and placed reversible tubal IUD and gavage analgesia at 4 weeks after operation. The control group 1 Counter-current method after modeling only placed reversible tubal IUD (distilled water after 4 weeks of gavage), control group 2 counter-current method 4 weeks after the operation with Huayu Zhitong scattered stomach, blank control group but not to model Distilled water gavage. Radioimmunoassay was used to detect the levels of OPN and HMGB1 in the serum of each group before modeling, after modeling and after treatment. Results Compared with the blank group (before model making), the levels of serum OPN and HMGB1 in the rats in each group were significantly increased (all P <0.05). The levels of serum OPN and HMGB1 in the experimental group and the control group were decreased, (P <0.05). The difference between the experimental group and the two control groups was also statistically significant (P <0.05). Conclusion OPN and HMGB1 play an important role in the development and progression of EM. Drug-loaded fallopian tube IJFHX may inhibit the proliferation of ectopic endometrium by regulating the levels of OPN and HMGB1, and promote the subsidence of inflammatory mass Good anti-inflammatory analgesic effect.
