目的:探讨通络益智散对脑缺血损伤大鼠海马亚区c-Fos蛋白表达的影响。方法:动物随机分为模型组、通络益智散组和尼莫地平组,尼莫地平组、通络益智散组分别灌胃给予尼莫地平0.001 g·kg-1,通络益智散1.57 g·kg-1,模型组给予蒸馏水。预防性灌胃给药7 d后,采用双侧颈总动脉缺血再灌注法制造大鼠脑缺血损伤模型,缺血再灌后2 h、4 h分别免疫组化法检测各组大鼠海马亚区c-Fos蛋白表达情况,并进行组间均数比较。结果:各组海马CA1、CA2、CA3、齿状回、下托区均可见c-Fos的阳性表达。与模型组比较,通络益智散、尼莫地平组海马各亚区c-Fos蛋白表达数量在造模后2h、4h明显降低(P<0.01)。通络益智散组与尼莫地平组比较无明显差异(P>0.05)。各组c-Fos蛋白数量在脑缺血损伤后2 h均达到高峰,4 h出现下降,对比有明显差异(P<0.01)。结论:通络益智散能够抑制脑缺血损伤c-Fos蛋白表达,具有神经保护作用。
Objective: To investigate the effects of Tongluo Yizhi Powder on c-Fos protein expression in hippocampal subfields of rats with cerebral ischemia injury. Methods: The animals were randomly divided into model group, Tonglu Yizhi powder group and nimodipine group, nimodipine group and Tongluo Yizhi powder group were given nimodipine 0.001 g · kg-1, Tongluo Yizhi Dissolve 1.57 g · kg-1, the model group given distilled water. After 7 days of prophylactic gavage, cerebral ischemia injury model was induced by bilateral common carotid artery ischemia-reperfusion in rats. Immunohistochemical method was used to detect the rats of each group 2 h and 4 h after ischemia reperfusion Hippocampus subregion c-Fos protein expression, and compared between groups. Results: The positive expression of c-Fos in all the hippocampal CA1, CA2, CA3, dentate gyrus and inferior centrum was observed. Compared with the model group, the expression of c-Fos protein in the hippocampus subgroups of Tonglu Yizhisan and nimodipine groups decreased significantly 2h and 4h after modeling (P <0.01). There was no significant difference between Tongluo Yizhi Powder group and nimodipine group (P> 0.05). The number of c-Fos protein in each group peaked at 2 h after cerebral ischemic injury and decreased at 4 h, with significant difference (P <0.01). Conclusion: Tongluo Yizhi San can inhibit the expression of c-Fos protein in cerebral ischemia injury and has neuroprotective effect.
