目的:利用近红外漫反射光谱分析技术对注射用丹参多酚酸中的水分进行无损、快速的定量分析。方法:以注射用丹参多酚酸为分析对象,用光纤测定近红外漫反射光谱,采用偏最小二乘法(PLS)建立定量校正模型。结果:53份样品的原始光谱经二阶导数预处理,选取11 993.1~6 098.8 cm-1和5 450.7~4 598.2 cm-1谱段,建立注射用丹参多酚酸(冻干)水分定量校正模型并分析,结果R2为0.991 1,内部交叉验证均方差(RMSECV)为0.210,最佳维数为4。10份样品用于外部验证,预测误差均方差(RMSEP)为0.243。结论:该方法分析快速、简便,结果准确可靠,可实现大批量样品的快速测定,对中药制剂的质量分析有一定的参考价值。
OBJECTIVE: To make a non-destructive and rapid quantitative analysis of water content in Salvia miltiorrhiza polyphenols for injection by near-infrared diffuse reflectance spectroscopy. Methods: Salvia miltiorrhiza polyphenols were used as the analysis object, the near-infrared diffuse reflectance spectra were measured by optical fiber, and the quantitative correction model was established by partial least squares (PLS). Results: The original spectra of 53 samples were pretreated with second order derivative. The spectral ranges of 11993.1 ~ 6098.8 cm-1 and 5 450.7 ~ 4 598.2 cm-1 were selected to quantify the water content of salvia miltiorrhiza The results showed that R2 was 0.991 1, root mean square error of internal cross validation (RMSECV) was 0.210, and the optimal dimension was 4.10 samples for external validation. The RMSEP was 0.243. Conclusion: The method is rapid, simple, accurate and reliable, and can be used for the rapid determination of large quantities of samples. It has certain reference value for the quality analysis of traditional Chinese medicine preparations.
