目的:研究丹参多酚酸盐及三七总苷对EA-hy926细胞在细胞增殖、血管新生及促进血管内皮生长因子(VEGF)表达方面的不同作用。方法:通过体外培养EA-hy926细胞,并给予不同浓度的丹参多酚酸盐和三七总苷(0.4,0.8和1.2mg·L–1)干预,运用CCK-8测定细胞增殖能力,培养12h后观察血管新生情况。培养24h后,通过real-timePCR方法观察VEGF的表达情况。结果:在浓度为0.4,0.8mg·L–1时,与空白对照比较,丹参多酚酸盐和三七总苷均能有效促进血管新生及VEGF的表达((P<0.05)),而两者比较无明显差异。在浓度为1.2mg·L–1时,与三七总苷相比,丹参多酚酸盐组VEGF的表达明显增强(P<0.05)。结论:丹参多酚酸盐和三七总苷均能有效促进EA-hy926细胞增殖、血管新生及VEGF的表达,而实验结果提示丹参多酚酸盐具有更强的促血管新生能力。
Objective: To study the different effects of Salviae miltiorrhizae polyphenols and Panax notoginseng saponins on cell proliferation, angiogenesis and the expression of vascular endothelial growth factor (VEGF) in EA-hy926 cells. Methods: EA-hy926 cells were cultured in vitro and treated with Salviae miltiorrhizae polyphenols and Panax notoginseng glycosides (0.4, 0.8 and 1.2 mg · L-1) at different concentrations. CCK-8 was used to determine the cell proliferation. After the observation of angiogenesis. After culturing for 24 hours, the expression of VEGF was observed by real-time PCR. Results: Salviae miltiorrhizae polyphenols and Panax notoginseng glycosides could effectively promote angiogenesis and VEGF expression at the concentrations of 0.4 and 0.8 mg · L -1 (P <0.05) No significant difference. At the concentration of 1.2 mg · L-1, the expression of VEGF in Salvia miltiorrhiza polyphenols group was significantly higher than that in Panax notoginseng glycosides (P <0.05). Conclusion: Salviae miltiorrhizae polyphenols and Panax notoginseng glycosides can effectively promote the proliferation, angiogenesis and VEGF expression of EA-hy926 cells, and the results suggest that Salvia miltiorrhiza polyphenols have stronger pro-angiogenic ability.
