目的:观察补阳还五汤对局灶性脑缺血大鼠脑组织APJ、Apelin、p-PI3K表达的影响,研究补阳还五汤的作用机制。方法:将成年大鼠随机分为正常对照组、假手术组、模型组和补阳还五汤组,采用大脑中动脉线栓法建立大脑中动脉阻塞模型,每组于给药后1、3、7 d处死,用Western blot及RT-PCR法检测各组不同时间点APJ、AKT、p-PI3K的蛋白及APT、Apelin mRNA表达。结果:补阳还五汤组各时间点APJ、p-PI3K、p-AKT蛋白及APT、Apelin mRNA表达显著高于模型组(P<0.05)。结论:补阳还五汤可减轻局灶性脑缺血后大鼠脑组织的损伤,其机制与调节缺血后脑组织APJ、Apelin、p-PI3K、p-AKT的表达有关。
Objective: To observe the effect of Buyang Huanwu Decoction on the expression of APJ, Apelin and p-PI3K in rat brain after focal cerebral ischemia and to study the mechanism of Buyang Huanwu Decoction. Methods: Adult rats were randomly divided into normal control group, sham operation group, model group and Buyang Huanwu Decoction group. Middle cerebral artery occlusion model was established by middle cerebral artery occlusion method. And then sacrificed at 7 days. The protein expression of APJ, AKT and p-PI3K and the expression of APT and Apelin mRNA were detected by Western blot and RT-PCR at different time points. Results: The expressions of APJ, p-PI3K and p-AKT protein and APT and Apelin mRNA in the Buyang Huanwu Decoction group were significantly higher than those in the model group at each time point (P <0.05). Conclusion: Buyang Huanwu decoction can reduce brain injury after focal cerebral ischemia in rats. Its mechanism is related to the regulation of the expression of APJ, Apelin, p-PI3K and p-AKT in ischemic brain tissue.
