目的通过对比美国药典,欧洲药典和中国药典检测大蒜(粉)功效成分的区别,理清国内外大蒜(粉)功效成分的研究方向,并对美国药典检测大蒜粉的功效成分——蒜氨酸的检测方法进行优化,提出一个操作更加简便,计算更加准确的蒜氨酸检测方法。方法称取试样适量用70%乙醇超声提取,进行高效液相色谱法分析,用十八烷基硅烷键合硅胶为填料的色谱柱分离,流动相为甲醇:5 mmol/L庚烷磺酸钠(磷酸调p H至2),蒜氨酸标准品(美国药典)定量计算其含量。结论蒜氨酸浓度在0.035616~0.213696 mg/m L的范围内与峰面积的线性良好,相关系数r>0.999。在80%、100%、120%添加水平下,蒜氨酸的回收率为98.0%。结论该方法操作简便、准确、重现性好,适用于大蒜粉中蒜氨酸的含量测定。
OBJECTIVE To compare the efficacy of different ingredients of garlic (powder) in the United States Pharmacopoeia, European Pharmacopoeia and Chinese Pharmacopoeia to clarify the research directions of the efficacy ingredients of garlic (powder) at home and abroad, and to study the efficacy of the United States Pharmacopoeia Optimization of detection methods, proposed a more simple operation, more accurate calculation of alliin detection methods. Methods The samples were weighed and extracted with 70% ethanol by ultrasonic and analyzed by high performance liquid chromatography with octadecylsilane bonded silica gel column. The mobile phase was methanol: 5 mmol / L heptane sulfonic acid Sodium (Phosphorus tune p H to 2), Alliin standard (United States Pharmacopoeia) Quantitative calculation of its content. Conclusions Alliin has good linearity with peak area in the range of 0.035616 ~ 0.213696 mg / m L with the correlation coefficient r> 0.999. The recoveries of alliin at 98% were 80%, 100% and 120%. Conclusion The method is simple, accurate and reproducible. It is suitable for the determination of alliin in garlic powder.
