目的综合运用目前较成熟的技术,建立一个易施行的、可对大组织标本连续切片整体图像进行三维重建及形态学测量的方法。方法对正常大鼠肾的石蜡包埋标本进行全序列连续切片,HE染色后对切片图像进行数字化采集,经过拼接、配准步骤,建立切片整体图像的数字化数据集,对其内部的血管系统及肾盂进行分割及三维重建,并对重建结果进行形态学测量。结果切片的整体图像清晰,数字化图像数据集配准准确,三维重建结果再现了各种结构的立体特征及其在原组织块内的位置,在三维空间内,可进行无阻挡的自由观察,并且测量出了脉管系统的多项三维形态学指标,包括组织体积构成百分比、动脉长度及分支角度等。结论采用这种方法取得了较为满意的重建结果 ,可为其他组织连续切片的三维重建研究提供借鉴。
OBJECTIVE To comprehensively use the currently more mature techniques to establish an easy-to-implement method for three-dimensional reconstruction and morphometric measurement of the whole slice of a large section of serial tissue sections. Methods Serial paraffin sections of paraffin-embedded specimens of normal rat kidneys were sectioned serially. After HE staining, the sections were digitally collected. After splicing and registration, the digitalized data sets of whole sections of the slices were established. The internal vascular system and The renal pelvis was segmented and reconstructed three-dimensionally, and the reconstruction results were measured morphologically. Results The whole image of the slice was clear and the registration of digital image dataset was accurate. The three-dimensional reconstructed results reproduced the stereoscopic features of various structures and their positions in the original tissue block. In the three-dimensional space, they were able to observe freely and without obstruction. The vascular system of a number of three-dimensional morphological indicators, including tissue volume percentage, length and branch angle and so on. Conclusion This method has achieved satisfactory reconstruction results, which can provide reference for the study of three-dimensional reconstruction of serial sections of other tissues.
