目的探讨神经递质NO、NPY在压力性尿失禁大鼠泌尿生殖道组织及耻尾肌中的表达及其与压力性尿失禁发生的关系。方法建立压力性尿失禁动物模型,取建模成功的SD雌性大鼠15只作为实验组,未经任何处理的正常大鼠20只作为对照组。处死后取尿道、阴道前壁及耻尾肌,HE染色,观察上述标本的组织形态学差异,并行免疫组化染色方法检测NO与NPY的分布及其表达差异。结果 HE染色显示,实验组大鼠的尿道、阴道前壁、耻尾肌与对照组相比,组织萎缩变薄,肌纤维排列疏松、紊乱。免疫组化染色显示,NO与NPY在大鼠尿道、阴道前壁、耻尾肌表达,与对照组相比,实验组表达减弱,两组相比差异有显著性(P<0.05)。结论 NO、NPY可能与压力性尿失禁发生有关。
Objective To investigate the expression of neurotransmitter NO and NPY in genitourinary tract tissues and shark muscle of stress urinary incontinence rats and their relationship with stress urinary incontinence. Methods The animal model of stress urinary incontinence was established. Fifteen SD female rats were selected as the experimental group and 20 normal rats without any treatment as the control group. The urethra, the anterior vaginal wall and the pubescent muscle were removed after sacrifice, and the histopathological changes were observed by HE staining. The distribution and expression of NO and NPY were detected by immunohistochemical staining. Results HE staining showed that compared with the control group, the urethra, anterior vaginal wall and the pubescent muscle in the experimental group showed atrophy and thinning, and the muscle fiber arranged loosely and disorderly. Immunohistochemical staining showed that the expression of NO and NPY in the urethra, anterior vaginal wall and shaman muscle of rats were significantly decreased compared with the control group, and there was significant difference between the two groups (P <0.05). Conclusion NO and NPY may be related to stress urinary incontinence.
