为研制一种 EB 病毒(EBV)基因探针,从鼻咽癌患者的活检组织、鼻咽分泌物和唾液中分离出 EBV,并从病毒中提取 DNA,克隆后用[α-~(32)P]dCTP 标记,制成[α-~(32)P]EBV-DNA 探针。再以患者活检标本、白细胞标本进行斑点杂交,用放射自显影法显示结果。临床试用表明:该探针诊断鼻咽癌的特异性强,灵敏度高,阳性率为92%(80/87),特异性为94.1%以上。这为鼻咽癌及与 EBV 有关疾病的诊断提供了一项检测方法,对上述疾病的流行病学调查及肿瘤基因学研究也有一定意义。
To develop an Epstein-Barr virus (EBV) gene probe, EBV was isolated from biopsies, nasopharyngeal secretions and saliva from patients with nasopharyngeal carcinoma and DNA was extracted from the virus. After cloning, P] dCTP tag to make [α- ~ (32) P] EBV-DNA probe. Then patient biopsy specimens, white blood cells specimens hybridization, autoradiography showed the results. Clinical trials show that: the probe diagnosis of nasopharyngeal carcinoma with strong specificity and high sensitivity, the positive rate was 92% (80/87), the specificity of 94.1%. This provides a detection method for the diagnosis of nasopharyngeal carcinoma and EBV-related diseases, and has certain significance for the epidemiological investigation and tumor gene research of the above diseases.
