目的通过研究黄芪注射液对实验性胰腺纤维化大鼠TGF-β1及其Smad2/3表达影响,探索其缓解胰腺纤维化的机制。方法大鼠随机分为5组,模型组和治疗组采用胰胆管注射2%TNBS溶液的方法制备慢性胰腺纤维化大鼠模型,治疗组分别给予低、中、高剂量的黄芪注射液治疗;造模后第6周,免疫组化检测胰腺TGF-β1、Smad2/3和Ⅰ型胶原的表达。结果与模型组相比,随着黄芪注射液剂量的增加,治疗组大鼠胰腺TGF-β1、Smad2/3和Ⅰ型胶原表达逐渐减少(P<0.05),具有统计学意义。结论黄芪注射液可以阻断TGF-β1的Smad2/3信号通路,缓解胰腺的纤维化。
Objective To study the effect of Astragalus injection on the expression of TGF-β1 and Smad2 / 3 in rats with experimental pancreatic fibrosis, and to explore its mechanism of relieving pancreatic fibrosis. Methods The rats were randomly divided into 5 groups. Chronic pancreatic fibrosis rat model was established by injecting pancreaticobiliary duct with 2% TNBS in the model group and treatment group. The treatment group was treated with Astragalus injection at low, medium and high doses respectively. At the 6th week, the expression of TGF-β1, Smad2 / 3 and type Ⅰ collagen in the pancreas were detected by immunohistochemistry. Results Compared with the model group, the expression of TGF-β1, Smad2 / 3 and type Ⅰ collagen in the pancreas decreased gradually with the increase of Astragalus injection (P <0.05), which was statistically significant. Conclusion Astragalus injection can block Smad2 / 3 signaling pathway of TGF-β1 and alleviate pancreatic fibrosis.
