目的 探讨低浓度的紫杉醇对人腺样囊性癌细胞株 ACC- 2粘附性及聚集血小板的影响。方法 采用MTT法分别检测 ,0 .1、 0 .0 6、 0 .0 1m g/L 的紫杉醇作用下 ,人腺样囊性癌细胞株 ACC- 2对细胞外基质 (ECM)的粘附和对血小板的聚集 ,并与无紫杉醇组对照。同时用 MTT法检测 ,0 .1、 0 .0 6、 0 .0 1m g/L紫杉醇对人腺样囊性癌细胞株 ACC- 2的细胞毒作用。结果 经紫杉醇处理后的 ACC- 2细胞株 ,粘附细胞外基质及聚集血小板的能力下调 ,并且和紫杉醇浓度呈剂量依赖关系 ,同时 ,此低浓度紫杉醇对 ACC- 2的细胞毒作用不明显。结论 紫杉醇能够明显抑制 ACC-2的粘附能力及聚集血小板的能力 ,并且与其细胞毒作用无关 ,因此紫杉醇具有抑制腺样囊性癌的侵袭及转移的潜能
Objective To investigate the effect of low concentration paclitaxel on the adhesion and aggregation of human adenoid cystic carcinoma cell line ACC-2. Methods MTT assay was used to detect the adhesion of ACC-2 to extracellular matrix (ECM) under the action of paclitaxel of 0.1, 0.06 and 0.01 mg / L respectively Platelet aggregation, and with no paclitaxel group control. At the same time, MTT assay was used to determine the cytotoxic effect of paclitaxel on human adenoid cystic carcinoma cell line ACC-2 at 0 .1, 0.06 and 0.01 m g / L. Results The paclitaxel-treated ACC-2 cell line had a decreased ability of adhering to the extracellular matrix and aggregating platelets, and the concentration of paclitaxel was dose-dependent. Meanwhile, the cytotoxic effect of paclitaxel at low concentration on ACC-2 cells was not obvious. Conclusion Paclitaxel can significantly inhibit the adhesion of ACC-2 and the ability of platelet aggregation, and has nothing to do with its cytotoxicity, so paclitaxel has the potential to inhibit the invasion and metastasis of adenoid cystic carcinoma
