目的:用多波长高效液相色谱法同时测定清胃黄连片中栀子苷、黄芩苷、盐酸小檗碱和甘草酸铵的含量。方法:采用phenomenex C18色谱柱(4.6 mm×250 mm,5μm),以乙腈(A)-0.5%三乙胺水溶液(B,以磷酸调pH值至3.0)为流动相梯度洗脱,检测波长分别为238 nm(栀子苷)、280 nm(黄芩苷)、348 nm(盐酸小檗碱)和250 nm(甘草酸铵)。结果:栀子苷、黄芩苷、盐酸小檗碱和甘草酸铵4种成分色谱峰分离符合要求;该4种成分加样回收率分别为102.5%(RSD=2.6%)、101.6%(RSD=2.9%)、102.6%(RSD=2.2%)和102.8%(RSD=2.1%);3批样品中栀子苷、黄芩苷、盐酸小檗碱和甘草酸铵每片平均含量分别为3.11 mg、14.66 mg、14.43 mg和8.04 mg。结论:利用多波长高效液相色谱法能同时测定清胃黄连片中4种有效成分的含量,结果准确、可靠。
Objective: To determine the content of geniposide, baicalin, berberine and ammonium glycyrrhizinate in Qingfeelian tablets by multi-wavelength high performance liquid chromatography. METHODS: The mobile phase was eluted with phenomenex C18 column (4.6 mm × 250 mm, 5 μm) with acetonitrile (A) -0.5% triethylamine (B, adjusted to pH 3.0 with phosphoric acid) 238 nm (geniposide), 280 nm (baicalin), 348 nm (berberine hydrochloride) and 250 nm (ammonium glycyrrhizinate). Results: The chromatographic peaks of geniposide, baicalin, berberine and ammonium glycyrrhizinate were satisfactory. The recoveries of these four components were 102.5% (RSD = 2.6%), 101.6% (RSD = 2.9%), 102.6% (RSD = 2.2%) and 102.8% (RSD = 2.1%). The average contents of geniposide, baicalin, berberine hydrochloride and ammonium glycyrrhizinate in each batch were 3.11 mg, 14.66 mg, 14.43 mg and 8.04 mg. Conclusion: The multi-wavelength high performance liquid chromatography can simultaneously determine the content of four effective components in the Qingfeelian tablets. The results are accurate and reliable.
