目的:模建人IL-6Rα的三维结构,为构建可拮抗IL-6作用的IL-6Rα的突变体提供理论依据。方法:以生长激素受体为模板蛋白,在Internet互联网络上利用Swis-Model蛋白同源结构模建工具进行IL-6Rα配基结合区的三维结构模建。结果与结论:得到了人IL-6Rα配基结合区的三维结构,并对模型进行了评价,确定了IL-6Rα上与异六聚体复合物一致的作用界面,为构建IL-6Rα的突变体提供了理论依据,并提示双突变体C277D/H280I可能既与IL-6具有高亲和力,又可拮抗IL-6的作用
OBJECTIVE: To construct a three-dimensional structure of human IL-6Rα and provide a theoretical basis for constructing a mutant of IL-6Rα that can antagonize the action of IL-6. METHODS: The three-dimensional structure of IL-6Rα ligand-binding domain was constructed by using the growth hormone receptor as a template protein on the internet through Swis-Model homology modeling tool. RESULTS AND CONCLUSION: The three-dimensional structure of the human IL-6Rα ligand binding region was obtained and the model was evaluated to determine the consistent interface of IL-6Rα with the hexameric complex for the construction of a mutation in IL-6Rα Provide a theoretical basis and suggest that the double mutant C277D / H280I may both have high affinity with IL-6 and antagonize IL-6
