小鼠骨髓细胞经程序降温-深低温保存后,给经致死剂量照射小鼠进行同种骨髓移植治疗造血障碍。实验用ICR.SMMC/C及昆明种小鼠219只,体重26~33g 8~9周龄,冻存骨髓细胞融解后,台盼蓝拒染率为70%~80%,CFU-GM,CFU-S测试均与新鲜骨髓结果相似。同种移植后动物活存率达43.6%,与新鲜骨髓无明显差别。冻存骨髓细胞移植加从小牛脾脏提取的抑素(Chalone)体内处理,动物活存率为42.5%,而Chalone体外处理活存只有25.0%,Chalone在冻存骨髓细胞移植中的作用尚待进一步实验。
Mice bone marrow cells were programmed to cool - cryopreservation, the lethal dose irradiated mice for allogeneic bone marrow transplantation for hematopoietic disorders. 219 ICR mice, Kunming mice and ICR mice were weighed 26-33 g for 8-9 weeks. The rate of trypan blue exclusion was 70% -80% after cryopreserved bone marrow cells were thawed. CFU-GM, CFU -S tests are similar to fresh bone marrow results. Animal survival rate after allograft reached 43.6%, with no significant difference between fresh bone marrow. Cryopreserved bone marrow cell transplantation plus in vivo treatment of calluses extracted from calves spleen extract, animal survival rate was 42.5%, while Chalone in vitro survival was only 25.0%, the role of Chalone in cryopreserved bone marrow cell transplantation needs further experiment.
