目的:建立HPLC同步测定天舒胶囊中天麻素、5-羟甲基糠醛、阿魏酸3种成分的含量。方法:Phenomenex LunaC18色谱柱(4.6 mm×250 mm,5μm),甲醇(A)-0.1%磷酸水溶液(B)梯度洗脱(0~10 min,5%~10%A;10~16 min,10%A;16~50 min,10%~60%A),流速1.0 mL·min-1,检测波长221 nm(天麻素)、283 nm(5-羟甲基糠醛)、322 nm(阿魏酸),柱温30℃。结果:天麻素、5-羟甲基糠醛、阿魏酸分别在5.61~299,3.31~212,4.63~247 mg·L-1呈良好的线性关系;天舒胶囊中3种成分的加样回收率分别为99.74%,102.5%,100.3%,RSD分别为0.84%,1.53%,0.49%。5批天舒胶囊市售样品中天麻素、5-羟甲基糠醛和阿魏酸的含量分别为2.70~3.65,2.07~2.67,1.95~2.10 mg·g-1。结论:测定天麻素、5-羟甲基糠醛和阿魏酸的含量能够有效地控制天舒胶囊的产品质量。
OBJECTIVE: To establish a HPLC simultaneous determination of three components of gastrodin, 5-hydroxymethylfurfural and ferulic acid in Tianshu capsule. METHODS: A gradient elution was performed on a Phenomenex Luna C18 column (4.6 mm × 250 mm, 5 μm) and a gradient of methanol (A) -0.1% phosphoric acid in water (0 to 10 min, 5% to 10% The detection wavelength was 221 nm (gastrodin), 283 nm (5-hydroxymethylfurfural), 322 nm (ferulic acid ), Column temperature 30 ℃.
