目的:探讨升清降糖合剂对RINm5F细胞的保护作用,为临床治疗糖尿病提供实验依据。方法:以STZ(链脲佐菌素)诱导建立胰岛细胞损伤模型;采用半仿生提取法提取升清降糖合剂有效成分(简称SQ);以不同浓度(100、200、400μg.mL-1按生药折算)的SQ作用于正常组和模型组细胞,以CCK-8法检测细胞生存活性并结合硝酸酶还原法测定细胞上清中NO含量。研究结果:经CCK-8法检测确定建立模型条件为0.8mmol/LSTZ干预RINm5F细胞24h;400μg/mL药物组能明显提高细胞活性(P<0.01),并减少细胞培养液中NO含量(P<0.01)。研究结论:升清降糖合剂对STZ导致胰岛细胞损伤有保护作用。
Objective: To investigate the protective effects of Shengqing Jiangtang Mixture on RINm5F cells and to provide experimental evidence for the clinical treatment of diabetes mellitus. Methods: The islet cell injury model was induced by STZ (streptozotocin); the active ingredient of Shengqing Jiangtang Mixture (SQ) was extracted by semi-bionic extraction method; the effects of different concentrations (100, 200, 400μg.mL- Crude drug conversion) of SQ role in the normal group and model group cells by CCK-8 method to detect cell viability and combined with nitrate reductase method to determine the content of NO in the cell supernatant. The results of CCK-8 assay confirmed that the established model was established by treating the RINm5F cells with 0.8 mmol / LSTZ for 24 h. 400 μg / mL of the drug significantly increased the cell viability (P <0.01) and decreased the NO content in the cell culture medium (P < 0.01). Conclusion: Shengqingjiangtang Mixture has a protective effect on STZ-induced islet cell injury.
