应用放射配基结合分析技术测定原代培养大鼠贮脂细胞维甲酸受体(RAR)含量及维甲酸(RA)对原代大鼠贮脂细胞RAR含量的影响;用放射免疫分析法测定RA,维生素A棕榈酯(RN)对原代培养大鼠贮脂细胞cAMP含量的影响。结果显示贮脂细胞RAR明显减少,RA作用48h,贮脂细胞RAR含量升高。原代培养的贮脂细胞经RA、RP作用48h后,细胞cAMP含量增加。结果提示RA调控细胞增殖、胶原蛋白合成是通过恢复贮脂细胞RAR含量,改变细胞信号系统,进而调控相应基因表达而实现的。
Radioligand binding assay was used to determine the content of retinoic acid receptor (RAR) and the content of retinoic acid (RAR) on primary cultured rat fat storage cells. The radioimmunoassay , Vitamin A palmitate (RN) on the cAMP content of primary cultured rat fat storage cells. The results showed that the RAR of storage fat cells was significantly reduced, and the RAR content of storage fat cells increased after 48 hours of RA treatment. Primary culture of fat-storing cells by RA, RP after 48h, the cell cAMP content increased. The results suggest that RA regulates cell proliferation and collagen synthesis by restoring the RAR content of lipid-storing cells, changing the cell signaling system, and then regulating the expression of the corresponding genes.
