目的:研究北美海蓬子不同极性溶剂提取物的体外抗肿瘤活性及其作用机制。方法:采用超声辅助提取法分别获得北美海蓬子正己烷、乙酸乙酯、正丁醇、甲醇和水的5种提取物,采用MTT法检测提取物对A549、MCF-7、Hela细胞毒作用,阳性对照为Adri。采用流式细胞术检测乙酸乙酯提取物对Hela细胞周期的影响。结果:正己烷、乙酸乙酯、正丁醇3种提取物对肿瘤细胞的生长抑制率达60%以上;乙酸乙酯提取物对Hela细胞的IC50可达17.6μg/mL,可使Hela细胞周期阻滞在G0/G1,显著减少G2/M期细胞比例。结论:北美海蓬子正己烷、乙酸乙酯、正丁醇提取物均对肿瘤细胞的体外增殖有抑制作用,其中乙酸乙酯提取物效果最强,可影响宫颈癌Hela细胞的细胞周期和促进细胞凋亡,其机制值得深入研究。
OBJECTIVE: To study the antitumor activity and mechanism of different polar solvent extract of S. japonicus in vitro. Methods: Five extracts of n-hexane, ethyl acetate, n-butanol, methanol and water were obtained by ultrasonic-assisted extraction. The cytotoxicity of extracts on A549, MCF-7 and Hela cells were detected by MTT assay , Positive control for Adri. The effect of ethyl acetate extract on Hela cell cycle was detected by flow cytometry. Results: The three kinds of extracts of n-hexane, ethyl acetate and n-butanol inhibited the growth of tumor cells by more than 60%. The IC50 of ethyl acetate extract reached 17.6μg / mL for Hela cells, Blocking at G0 / G1 significantly reduced the proportion of G2 / M phase cells. CONCLUSION: The n-hexane, ethyl acetate and n-butanol extract of S. japonicus have inhibitory effects on the proliferation of tumor cells in vitro. The ethyl acetate extract has the strongest effect on the cell cycle and the promotion of cervical cancer Hela cells Apoptosis, the mechanism worth in-depth study.
