目的:研究复方加味温胆汤中的挥发性成分,探讨其治疗自闭症及相关症状的药效物质基础。方法:分别采用顶空固相微萃取法(HS-SPME)、水蒸气蒸馏法(SD)结合气质联用技术(GC-MS)对复方中挥发性成分进行分析鉴定;采用HS-SPME-GC-MS对煎煮前后复方挥发性成分进行对比分析。结果:共鉴定复方挥发性成分187种,含量最高的为α-细辛醚,D-柠檬烯次之。采用SD-GC-MS检测出74种,鉴定出66种,占挥发性成分总量99.89%;采用HS-SPME-GC-MS从煎煮前复方中检测出挥发性成分104种,鉴定出85种,占总挥发性成分的99.79%,从煎煮后复方中检测出挥发性成分77种,鉴定出65种,占总挥发性成分95.45%。结论:复方中α-细辛醚、D-柠檬烯和右旋龙脑等5种挥发性成分具有调节中枢神经系统、改善大脑功能、抗惊厥和安定等作用,能够改善自闭症的症状。本研究为复方加味温胆汤的挥发性成分分析提供了方法学基础,同时也为其治疗自闭症药效物质基础研究提供了参考依据。
OBJECTIVE: To study the volatile components in Compound Modified Wendan Decoction and to explore its pharmacodynamic basis for the treatment of autism and related symptoms. Methods: The volatile components in the compound were identified by headspace solid-phase microextraction (HS-SPME) and steam distillation (SD) combined with gas chromatography-mass spectrometry (GC-MS) -MS comparative analysis of volatile components before and after decoction. Results: A total of 187 volatile components were identified, the highest content of α-asarone, followed by D-limonene. 74 species were identified by SD-GC-MS, 66 species were identified, accounting for 99.89% of the total volatile components; 104 volatile components were detected by HS-SPME-GC-MS before decoction, 85 Species, accounting for 99.79% of the total volatile components, 77 compounds were detected from the compound after boiling, 65 kinds were identified, accounting for 95.45% of the total volatile components. CONCLUSION: Five kinds of volatile components such as α-asarone, D-limonene and dexamethasone can improve central nervous system, improve brain function, anticonvulsant and diazepam, and improve the symptoms of autism. This study provides a methodological basis for the analysis of the volatile components of the compound Wendan Wendan Decoction, and also provides a reference for its basic research on the therapeutic substances of the drug of autism.
