目的 建立测定青娥丸提取物中松脂醇二葡萄糖苷含量的RP HPLC法。方法 青娥丸提取物用 75 %乙醇提取 ,经AB 8大孔吸附树脂纯化后 ,注入高效液相色谱仪。本法采用ODS分析柱为固定相 ,以甲醇 乙腈 水 (2 4∶3∶78,V V V)为流动相 ,流速为 1 0mL·min- 1 ,紫外检测波长为 2 2 7nm。结果 松脂醇二葡萄糖苷的线性范围为 5 5 170 μg·mL- 1 (r>0 9998) ,平均回收率 99 3%。日内和日间精密度分别为 1 3%和 2 8% (n =5 )。青娥丸中松脂醇二葡萄糖苷的含量为 0 4 4 6± 0 0 12mg·g- 1 (n =10 )。结论 本方法灵敏、准确 ,专属性强 ,适用于青娥丸及其他复方中药中松脂醇二葡萄糖苷的含量测定
Objective To establish a RP HPLC method for the determination of pinoresinol diglucoside in Qingyangwan extract. Methods Qing’e pill extract was extracted with 75% ethanol, purified by AB 8 macroporous resin and injected into high performance liquid chromatography. ODS analytical column was used as the stationary phase in this method. The mobile phase consisted of methanol and acetonitrile (2: 3: 78, V V V) with flow rate of 10 mL · min-1 and UV detection wavelength of 227 nm. Results The linear range of pinoresinol diglucoside was 55 170 μg · mL -1 (r> 0 9998) with an average recovery of 99.3%. Intra- and inter-day precision was 13% and 28%, respectively (n = 5). The content of pinoresinol diglucoside in Qing’e pills was 0 446 ± 0 0 12 mg · g -1 (n = 10). Conclusion The method is sensitive, accurate and specific and is suitable for the determination of pinoresinol diglucoside in Qing’e pills and other compound Chinese herbs
