目的探讨氧化应激在糖尿病肌病发生发展中的作用。方法尾静脉内注射链脲佐菌素诱导糖尿病大鼠模型。测定糖尿病不同时期股四头肌组织丙二醛含量和总超氧化物歧化酶活性。光镜下观察糖尿病不同时期股四头肌组织形态学的改变。结果糖尿病大鼠1,3和6个月股四头肌组织丙二醛水平与同期对照组相比显著增高,分别为(1.81±0.30),(0.96±0.35),P<0.01;(1.14±0.20)(0.91±0.21),P<0.05;(1.32±0.29),(0.97±0.22),P<0.05。糖尿病大鼠1个月股四头肌组织总超氧化物歧化酶活性与对照组相比明显增高(47.43±6.23),(34.21±4.04),P<0.01,3个月时降至正常对照水平,6个月时明显降低(26.10±5.89),(32.67±5.56),P<0.05。组织形态学观察表明:糖尿病大鼠3个月时股四头肌出现病理改变,随着病程延长,病理改变越严重。结论糖尿病大鼠早期股四头肌就已存在明显的氧化应激,氧化应激在糖尿病肌病的发生发展中起重要作用。
Objective To investigate the role of oxidative stress in the development of diabetic myopathy. Methods The model of streptozotocin-induced diabetic rats was induced by tail vein injection. The malondialdehyde (MDA) content and total superoxide dismutase (SOD) activity of quadriceps femoris in different periods of diabetes mellitus were measured. Light microscopic observation of diabetic quadrats during different periods of histological changes. Results Compared with the control group, the level of malondialdehyde in 1,3 and 6 months quadriceps muscle of diabetic rats were significantly increased (1.81 ± 0.30), (0.96 ± 0.35), P <0.01 (1.14 ± 0.20) (0.91 ± 0.21), P <0.05; (1.32 ± 0.29), (0.97 ± 0.22), P <0.05. Compared with the control group, the total superoxide dismutase activity in quadriceps muscle of diabetic rats increased significantly (47.43 ± 6.23), (34.21 ± 4.04), P <0.01, and decreased to the level of normal control , Significantly lower at 6 months (26.10 ± 5.89), (32.67 ± 5.56), P <0.05. Histopathological examination showed that pathological changes of quadriceps femoris occurred in diabetic rats at 3 months, and the pathological changes became more serious with the prolongation of the course. Conclusion Early oxidative stress exists in the early quadriceps femoris of diabetic rats, and oxidative stress plays an important role in the development of diabetic myopathy.
