目的:探讨冠脉通片对大鼠血栓形成的保护作用及其机制。方法:将SD大鼠随机分成5组:模型组(M)、假手术组(S)、冠脉通低剂量组(GL)、冠脉通中剂量组(GM)和冠脉通高剂量组(GH),体外动脉血栓形成方法构建血栓形成模型并检测各组间血栓湿重的差异,通过检测凝血时间和凝血相关因子[P-选择素和血管性血支因子(v WF)]的表达情况反应冠脉通片保护大鼠血栓形成的作用机制。结果:GL、GM和GH组与M组大鼠比较,血栓湿重降低(P<0.05),GH组作用最明显,与此同时血栓抑制率有所增加。M组凝血酶时间有所减少,GL、GM组活化部分凝血活酶时间(APTT)中与M组比较,差异有统计学意义(P<0.05);凝血酶原时间(PT)在各组间比较,差异无统计学意义(P>0.05)。P-选择素和v WF-1是血小板聚集及活化的重要因子,与M组比较,GL、GM组P-选择素和v WF-1水平较低(P<0.05)。结论:冠脉通片可通过影响凝血相关因子的表达对大鼠血栓形成起到保护作用。
Objective: To investigate the protective effect and mechanism of coronary vein tablet on thrombosis in rats. M group thrombin time decreased, GL, GM group activated partial thromboplastin time (APTT) compared with M group, the difference was statistically significant (P <0.05); prothrombin time (PT) The difference was not statistically significant (P> 0.05). P-selectin and v WF-1 were important factors of platelet aggregation and activation. Compared with M group, the levels of P-selectin and v WF-1 in GL and GM groups were lower (P <0.05). Conclusion: Guan Tong Tong Tablet can protect thrombosis in rats by affecting the expression of coagulation related factors.
