目的从肺腺癌A549细胞对拉伸应力响应时释放出的细胞形变调整行为,解读肿瘤细胞骨架异常与其生物学行为的相关性。方法使用单向等轴应变加载装置对培养的肺腺癌A549细胞施以周期性基底拉伸加载,通过计算机图像处理并与对照组进行比较。结果(1)A549细胞的形态在加载中有明显的变化,铺展面积明显增大,细胞外形变得非常不规则,即,细胞不规则度增加;(2)与对照组比较,加载组细胞细胞核的铺展面积增大,拉伸引起细胞核形态的变化比较小,加载组与对照组细胞核的形态差别不大;(3)周期性拉伸引起了细胞取向的调整,细胞调整到与主应变近似垂直的方向,许多细胞拉长首尾连成一线,线的取向趋于与主应变方向垂直。结论A549细胞的细胞骨架各组份间对应力的响应协同反应下降;肺腺癌A549细胞对应力方向的呼应尚属正常。
Objective To modulate the behavior of cell deformities released from lung adenocarcinoma A549 cells in response to tensile stress, and to interpret the correlation between cytoskeleton abnormalities and their biological behavior. Methods The cultured lung adenocarcinoma A549 cells were subjected to periodic basal tensile loading using a uniaxial equiaxed strain loading device. The image was processed by computer and compared with the control group. Results (1) The morphology of A549 cells changed significantly during loading, the area of spreading significantly increased, and the cell appearance became very irregular, ie, the irregularity of cells increased; (2) Compared with the control group, the cell nuclei of the loading group were compared with the control group. The spreading area increases, stretching causes the change of the nucleus morphology is relatively small, the morphology of the nucleus of the loading group and the control group has little difference; (3) The periodic orientation causes the adjustment of the cell orientation, and the cells are adjusted to be approximately perpendicular to the main strain. In the direction of many, many cells stretched in a straight line from end to end, and the orientation of the lines tends to be perpendicular to the main strain direction. CONCLUSION: The response to stress of A549 cells decreases in response to synergistic responses between the various components of the cytoskeleton; the response of the lung adenocarcinoma A549 cells to stress is normal.
