在80~90d日龄C57BL/6J小鼠上观察了10~1000μg/kgd-14种剂量rhIL-6对6.5Gyγ线(132~127.7R/min)照射后外周血细胞恢复的影响;观察时间持续至照射后30天。结果表明:白细胞、红细胞和血小板计数的动态变化时间进程给药组和对照组相似,但总的趋势表明给药组各种血细胞计数的最低值高于对照组、各时间点的恢复情况均不同程度地优于对照组;除10μg组外均有一部份时间点统计学差别显著,而500和1000μg剂量组效果尤为显著。血红蛋白含量和RBC比积的变化与RBC数一致。相关系数分析结果表明在一部分时间点上,外周血细胞数有明显的药量依赖性。照后22天WBC数、14和18天RBC数以及11和18天PLT数的剂量效应回归方程均具有统计学意义。本文结果表明:皮下注射rhIL-6剂量达到200μg/kgd-1以上连续4d可减轻6.5Gy照射小鼠外周血细胞数下降程度并促进其恢复。其机制可能是通过IL-6对造血干、祖细胞增殖与分化的改善而起作用。
The effects of 10 ~ 1000μg / kg d-14 doses of rhIL-6 on the recovery of peripheral blood cells after 6.5Gyγ line irradiation (132 ~ 127.7R / min) were observed in 80-90 days old C57BL / 6J mice. The regression equations of the dose-effect of WBC numbers, RBC counts on the 14th and 18th days and PLT numbers on the 11th and 18th days after 22 days were statistically significant. The results of this study show that: the subcutaneous injection of rhIL-6 dose of 200μg / kgd-1 more than 4d consecutive days can reduce the 6.5Gy irradiation mice peripheral blood cells decreased and promote its recovery. The mechanism may be through IL-6 hematopoietic stem and progenitor cell proliferation and differentiation play a role.
