目的 :观察IL 18对慢性乙肝患者Th1/Th2细胞分化的影响。方法 :分离 4 0例慢性乙型肝炎患者外周血单个核细胞 (peripheralbloodmonoclearcells,PBMC) ,分别与植物血凝素 (PHA ,10 0mg/L)、HBcAg(1mg/L)、HBeAg(1mg/L)单独或联合IL 18(10 μg/L)体外培养 4 8h ,ELISA法检测培养液中IL 2、IFN γ、IL 4、IL 10水平。 2 0例健康人群作对照。结果 :IL 18对健康人群PBMC产生的Th1/Th2类细胞因子无明显影响 ,但对慢性乙肝患者在IL 18和HBcAg或IL 18和HBeAg联合诱导下则显著增强PBMC产生的Th1类细胞因子IL 2和IFN γ ,但对Th2类细胞因子IL 4和IL 10无明显影响。结论 :IL 18增强慢性乙肝患者Th1类细胞因子的优势表达 ,从而促进Th1细胞的优势分化 ,而对Th2类细胞因子的表达无影响
Objective: To observe the effect of IL 18 on Th1 / Th2 cell differentiation in chronic hepatitis B patients. Methods: Peripheral blood mononuclear cells (PBMC) from 40 patients with chronic hepatitis B were isolated and compared with those of phytohemagglutinin (PHA), HBcAg (1mg / L) and HBeAg (1mg / L) The cells were incubated with IL-18 (10 μg / L) alone or in combination for 48 h. The levels of IL 2, IFN γ, IL 4 and IL 10 in the culture medium were detected by ELISA. Twenty healthy controls were used as controls. RESULTS: IL-18 had no significant effect on Th1 / Th2 cytokines produced by PBMC in healthy subjects, but IL-18 induced by PBMCs induced by IL-18 and HBcAg or IL-18 combined with HBeAg significantly increased IL-2 And IFNγ, but had no significant effect on Th2 cytokines IL4 and IL10. CONCLUSION: IL-18 enhances the predominant expression of Th1 cytokines in patients with chronic hepatitis B, thereby promoting the predominant differentiation of Th1 cells but not affecting the expression of Th2 cytokines
