目的:寻求能有效防治糖尿病神经病变的最佳中药复方制剂。方法:采用L18(37)正交表,考查三因素(葛根提取物、黄芪提取物和丹参提取物)在三水平(20mg/L,100mg/L,200mg/L)上预处理对低浓度(5mmol/L)过氧化氢(H_2O_2)致大鼠脑皮质线粒体损伤SOD酶活性的保护作用;考查三因素(葛根提取物、黄芪提取物和丹参提取物)在三水平(20mg/L,40mg/L,100mg/L)上预处理对高浓度(300mmol/L)H_2O_2致大鼠皮质脑线粒体损伤Na~+,K~+-ATP酶活性的保护作用。结果:经5mmol/L或300mmol/LH_2O_2处理的线粒体,其SOD活力以及Na~+,K~+-ATP酶活性均显著降低,与对照组比较有统计学差异。线粒体SOD活力分析结果显示:黄芪提取物和丹参提取物在增加SOD活性方面发挥主要作用,葛根*黄芪、葛根*丹参以及葛根*黄芪*丹参之间存在显著的协同增强作用。线粒体Na~+,K~+-ATP酶活性分析结果显示:黄芪提取物和丹参提取物在增加Na~+,K~+-ATP酶活性方面发挥主要作用,并且葛根*黄芪*丹参之间存在显著的协同增强作用。结论:由葛根提取物,黄芪提取物和丹参提取物以1:1:1剂量比例组成的中药复方制剂对H_2O_2所致大鼠脑线粒体损伤具有最佳保护作用。
Objective: To seek the best traditional Chinese medicine compound preparation that can effectively prevent and treat diabetic neuropathy. Methods: The three factors (Pueraria lobata extract, Astragalus membranaceus extract and Salvia miltiorrhiza extract) were pretreated on the three levels (20mg / L, 100mg / L and 200mg / L) (20 mg / L, 40 mg / L) on the activity of SOD in cortex mitochondria induced by hydrogen peroxide (H 2 O 5 mmol / L) L, 100mg / L) on the mitochondrial Na ~ +, K ~ + -ATPase activity in rat cerebral cortex with high concentration (300mmol / L) Results: The mitochondria treated with 5mmol / L or 300mmol / L H2O2 significantly decreased the activity of SOD and the activities of Na ~ + and K ~ + -ATPase, compared with the control group. The results of mitochondrial SOD activity showed that Astragalus membranaceus and Salvia miltiorrhiza extract had a significant role in increasing SOD activity. There was a significant synergistic enhancement effect between Radix Puerariae, Radix Puerariae Radix * and Radix Puerariae Radix * Radix Salviae Miltiorrhizae. Analysis of mitochondrial Na ~ +, K ~ + -ATPase activity showed that astragalus extract and Salvia miltiorrhiza extract play a major role in increasing Na ~ +, K ~ + -ATPase activity, and Pueraria * Radix Astragali, Significant synergistic effect. Conclusion: The traditional Chinese medicine preparation composed of Radix Puerariae extract, Astragalus extract and Radix Salviae Miltiorrhizae extract in a ratio of 1: 1: 1 has the best protective effect on mitochondrial damage induced by H_2O_2 in rats.
