目的探讨黄芪多糖对高同型半胱氨酸血症(Hhcy)大鼠心肌缺血再灌注损伤的保护作用及机制。方法应用腹腔注射2%L-半胱氨酸5mL/kg复制Hhcy大鼠模型。10周后,腹腔注射戊巴比妥钠麻醉下气管插管安装呼吸机,开胸结扎冠状动脉左室降支90分钟后松开结扎线再灌注90分钟。标Ⅱ导联全程记录心电图,并于再灌注90分钟取血。检测肌酸激酶(CK)、乳酸脱氢酶(LDH)、心肌肌钙蛋白Ⅰ(Tn-Ⅰ)、同型半胱氨酸(Hcy)。结果每天腹腔注射2%L-半胱氨酸5mL/kg,10周后,血浆Hcy升高。黄芪多糖干预组的CK、LDH、Tn-Ⅰ水平明显低于Hhcy模型对照组。结论黄芪多糖对Hhcy大鼠心肌缺血再灌注损伤具有保护作用。
Objective To investigate the protective effect of astragalus polysaccharides on myocardial ischemia reperfusion injury in hyperhomocysteinemia (Hhcy) rats and its mechanism. Methods Hhcy rat model was established by intraperitoneal injection of 2% L-cysteine 5mL / kg. Ten weeks later, intraperitoneal injection of sodium pentobarbital anesthesia intubation ventilator installed, thoracotomy coronary artery left ventricular descending artery 90 minutes after the release of ligature reperfusion for 90 minutes. Standard Ⅱ leads recorded the entire ECG, and 90 minutes after reperfusion blood. The creatine kinase (CK), lactate dehydrogenase (LDH), cardiac troponin Ⅰ (Tn-Ⅰ) and homocysteine (Hcy) were detected. Results Intraperitoneal injection of 2% L-cysteine 5mL / kg, after 10 weeks, plasma Hcy increased. Astragalus polysaccharide intervention group CK, LDH, Tn-Ⅰ levels were significantly lower than the Hhcy model control group. Conclusion Astragalus polysaccharide has a protective effect on myocardial ischemia-reperfusion injury in Hhcy rats.
