目的 :研究人胚肺 (HEL)细胞HOXB2 ,HOXB3,HOXB4及HOXB8基因的表达状态及人类巨细胞病毒(HCMV)感染对HEL细胞HOXB2 ,HOXB3,HOXB4及HOXB8基因表达的影响。方法 :采用半定量逆转录 -聚合酶链反应 (RT -PCR)技术。结果 :HEL细胞表达HOXB2 ,HOXB3,HOXB4及HOXB8;HOXB2基因于HCMV感染后 15h表达增加 ,4 8h达高峰 ,随后回落 ,至 12 0h再达高峰。HEL细胞HOXB3和HOXB4基因的表达在HCMV感染后不同时间无明显变化。HOXB8基因于HCMV感染后 15~ 4 8h表达轻微下调 ,72h回升 ,96h表达增加 ,12 0h达高峰 ;ATRA能轻微上调HCMV感染晚期HEL细胞HOXB8基因的表达 ,但对HCMV感染后HOXB2和HOXB4基因的表达无明显调节作用。结论 :HCMV能诱导HOXB2及HOXB8基因表达异常 ,这可能在先天性HCMV感染导致胚胎发育畸形中起重要作用。
Objective: To investigate the expression of HOXB2, HOXB3, HOXB4 and HOXB8 in human embryo lung (HEL) cells and the effect of human cytomegalovirus (HCMV) infection on the expression of HOXB2, HOXB3, HOXB4 and HOXB8 in HEL cells. Methods: Semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) was used. Results: The expression of HOXB2, HOXB3, HOXB4 and HOXB8 were detected in HEL cells. The expression of HOXB2 increased 15 h after HCMV infection and peaked at 48 h, then dropped back to the peak at 120 h. The expression of HOXB3 and HOXB4 genes in HEL cells showed no significant changes at different times after HCMV infection. The expression of HOXB8 gene was slightly downregulated 15 h to 48 h after HCMV infection and rose at 72 h, increased at 96 h, and peaked at 12 h. ATRA slightly up-regulated HOXB8 gene expression in HCMV-infected HEL cells. However, No significant role in the regulation of expression. Conclusion: HCMV can induce abnormal gene expression of HOXB2 and HOXB8, which may play an important role in embryonic developmental deformity caused by HCMV infection.
