目的 :分析能合成端粒DNA重复序列的端粒酶在DMBA诱导金黄地鼠颊囊癌变动态过程中的动态变化。方法 :采用PCR基础上的端粒DNA重复序列扩增法和ELISA酶免疫技术 ,测定颊囊癌变组织端粒酶动态变化。结果 :在诱癌过程中 ,颊囊粘膜组织端粒酶活性随组织病理学改变向恶性转化而增高 ,与自身对照相比较有显著差异 ,其升高在诱癌后期最明显。结论 :端粒酶激活是肿瘤形成的早期分子标志 ,也是端粒长度稳定的主要因素。
OBJECTIVE: To analyze the dynamic changes of telomerase, which can synthesize telomeric repeat sequences, in the process of carcinogenesis of golden hamster cheek pouch induced by DMBA. Methods: The telomerase DNA repeat amplification and enzyme-linked immunosorbent assay (ELISA) were used to determine the telomerase activity in the cheek pouch. Results: The telomerase activity of cheek pouch mucosa increased with pathological changes to malignant transformation in the process of cancer induction, which was significantly different from the control. Conclusion: Telomerase activation is an early molecular marker of tumor formation and is also the main factor of telomere length stability.
