本实验选用了雄性、成年、健康家兔68只。在阿米妥钠溶液静脉注射之麻醉情况下进行无菌手术:在挠骨中段人工造成4毫米的缺损。手术后1, 2, 3,,、7, 10, 14, 17,21, 28, 35及42天后杀死动物,取材,固定,进行了组织学和组织化学研究。 在骨折修复过程中,不同时期内可以见到三种不同的骨痴:(1)粗纤维编织状骨,(2)细纤维板层状骨,以及(3)混合性骨小梁。 当扁平的成骨细胞分化成高柱状的成骨细胞时,细胞内所包含的核糖核酸、碱性磷酸酶、细胞色素氧化酶及墟拍酸脱氢酶等逐渐增加。在成骨细胞分化成骨细胞过程中则上述的各种组织化学反应弦度逐渐减低。在细纤维骨小梁内的骨细胞,上述各种成分几乎完个消失;在粗纤维骨小梁的细胞内部仍旧显示了中等程度的上述各种组织化学反应。在新生的粗纤维骨小梁内骨基质和骨纤维内高碘酸雪夫氏反应是要较细纤维骨内的反应要强一些。在上述各种骨小梁的基质内,除了细纤维骨骨陷窝壁和粘合线内,都没有见到明显的酸性粘多糖反应和异染性;碱性磷酸酶的反应也仅限于骨外衣下粗纤维外包裹的细纤维。 本文尚结合破骨细胞的形态结构及组织化学反应讨论了破骨细胞的机能问题。
The experiment selected 68 male, adult, healthy rabbits. Aseptic surgery was performed under intravenous anesthesia with sodium amylate solution: a 4 mm defect was manually created in the middle of the radius. After 1, 2, 3, 7, 10, 14, 17, 21, 28, 35 and 42 days after operation, the animals were killed, drawn and fixed for histological and histochemical studies. In the process of fracture repair, three different types of bizarre can be seen in different periods: (1) crude fiber braided bone, (2) fine fibril lamellar bone, and (3) mixed trabecular bone. When flattened osteoblasts differentiate into highly columnar osteoblasts, the content of ribonucleic acid, alkaline phosphatase, cytochrome oxidase and phenylalanine dehydrogenase in the cells gradually increases. In the process of osteoblast differentiation into osteoblasts, the chord of various histochemical reactions described above gradually decreases. In the fine-fiber trabecular bone within the cells, the above-mentioned various components disappear almost completely; inside the crude fiber trabecular cells still showed a moderate degree of the above-mentioned various histochemical reactions. In the nascent crude fiber trabecular bone matrix and bone acid within the periodic acid Schiff’s response is to fine fibrocartilage reaction stronger. No significant acid mucopolysaccharide reaction and metachromaticity were seen within the matrix of the various trabecular planes except fine fibrocartilage lacunas and adhesive lines; alkaline phosphatase was also limited to bone Under the coat of coarse fibers wrapped thin fibers.
