目的观察人参皂苷Re对H9c2心肌细胞色素P450酶的影响,旨在发现人参皂苷Re对心肌细胞作用的分子机制。方法实验分对照组、人参皂苷Re各剂量组(1、5、10、50、100μmol·L~(-1))处理组,人参皂苷Re作用6、24、36、48及60 h后提取RNA或蛋白进行测定。利用实时定量PCR法检测H9c2心肌细胞CYP2C11、CYP2J3、CYP4A1、CYP4A3、CYP4F4及ANP mRNA的表达;采用Western blot法检测心肌细胞CYP4A1和CYP2J3蛋白的表达。结果人参皂苷Re明显上调心肌细胞CYP2C11、CYP2J3 mRNA的表达至正常对照的1.6、1.8倍,明显下调CYP4A1、CYP4A3及CYP4F4 mRNA的表达至正常对照的0.4、0.15、0.3倍。人参皂苷Re明显上调ANP基因表达水平至正常对照的3.2倍。随着药物浓度的增加,人参皂苷Re对CYP4A1蛋白表达产生明显的下调作用,而对CYP2J3蛋白产生明显的上调作用。结论人参皂苷Re可影响心肌细胞的CYP450酶和ANP基因的表达。
Objective To observe the effect of ginsenoside Re on H9c2 cardiomyocyte cytochrome P450 enzyme and to find out the molecular mechanism of ginsenoside Re on cardiomyocytes. Methods The experimental group, Ginsenoside Re treatment groups (1,5,10,50,100μmol·L -1) treatment group, ginsenoside Re at 6,24,36,48 and 60 h after RNA extraction Or protein. The expression of CYP2C11, CYP2J3, CYP4A1, CYP4A3, CYP4F4 and ANP mRNA in H9c2 cardiomyocytes was detected by real-time quantitative PCR. The expression of CYP4A1 and CYP2J3 protein in cardiomyocytes was detected by Western blot. Results Ginsenoside Re significantly up-regulated the expression of CYP2C11 and CYP2J3 mRNA in cardiomyocytes to 1.6 and 1.8 times of the normal control, and down-regulated the expression of CYP4A1, CYP4A3 and CYP4F4 mRNA to 0.4, 0.15 and 0.3 times of the normal control, respectively. Ginsenoside Re significantly increased ANP gene expression levels to 3.2 times the normal control. With the increase of drug concentration, ginsenoside Re significantly reduced the expression of CYP4A1 protein and upregulated CYP2J3 protein. Conclusion Ginsenoside Re can affect the expression of CYP450 enzyme and ANP gene in cardiomyocytes.
