目的:研究7-二氟甲氧基-5,4′-二-正辛烷氧基金雀异黄素(DFOG)抑制人肺癌细胞生长作用及机制。方法:用不同浓度DFOG处理体外培养A549细胞后,流式细胞术(FCM)检测细胞周期分布;平皿集落形成法测定细胞生长;Western blot分析FoxM1蛋白表达。结果:DFOG处理24 h导致A549细胞系G2期细胞百分率显著增加(P<0.05);DFOG以剂量依赖方式抑制A549细胞集落形成率(P<0.05)。Western Blot分析发现,2.0μmol/L DFOG以时间依赖方式下调A549细胞FoxM1表达。结论:DFOG抑制A549细胞生长与其降低FoxM1表达有关。
AIM: To investigate the inhibitory effects of DFOG on the growth of human lung cancer cells and its mechanism. Methods: A549 cells were treated with different concentrations of DFOG in vitro. Cell cycle distribution was detected by flow cytometry (FCM). Cell colony formation was measured by plate colony formation assay. FoxM1 protein expression was analyzed by Western blot. Results: DFOG treatment at 24 h resulted in a significant increase of G2 cell percentage in A549 cell line (P <0.05). DFOG inhibited A549 cell colony formation rate in a dose-dependent manner (P <0.05). Western Blot analysis showed that 2.0μmol / L DFOG down-regulated FoxM1 expression in A549 cells in a time-dependent manner. Conclusion: DFOG inhibits the growth of A549 cells and reduces the expression of FoxM1.
