目的 了解血小板衍化内皮细胞生长因子 (PD ECGF)在肝癌中的表达 ,探讨与其相关的卡培他滨 (capecitabine ,CAP)治疗对肝癌生长和转移的作用。方法 用免疫组织化学法检测 6 1例肝细胞癌 (HCC)患者肝组织中PD ECGF蛋白水平。应用NorthernBlot法对其中 2 8例HCC组织中PD ECGFmRNA水平作相对定量分析。用 2 4只裸鼠人肝癌高转移模型LCI D2 0 ,于肿瘤种植后第 3天分别采用不同剂量CAP治疗。停药后第 3天处死裸鼠 ,测量肿瘤的长短径 ,并用HE染色检测肺转移。结果 6 1例HCC和癌周组织中PD ECGF蛋白表达阳性率分别为 70 .5 %和 47.5 % ,37例高TNM分期组 (Ⅲ期和Ⅳ期 )HCC组织中PD ECGF蛋白表达阳性率 (81.1% )明显高于 2 4例低TNM分期组 (Ⅰ期和Ⅱ期 )表达率 (5 4.2 % ) ,两者差异有显著意义 (P <0 .0 5 )。NorthernBlot分析 ,HCC组织的PD ECGF蛋白水平表达与其相应mRNA水平变化一致。CAP 2 .10mmol·kg-1·day-1和 3.15mmol·kg-1·day-1治疗组裸鼠肝癌体积 (2 40mm3± 119mm3 和 2 0 9mm3± 15 0mm3)明显小于对照组 (4 47mm3± 15 9mm3) (P<0 .0 5 )。裸鼠肺转移率随CAP剂量的增加而下降 (P <0 .0 5 )。结论 PD ECGF在HCC组织中高表达 ,与肝癌TNM分期相关。CAP能抑制HCC的生长和转移。
Objective To investigate the expression of platelet-derived endothelial cell growth factor (PD-ECGF) in hepatocellular carcinoma (HCC) and to investigate the effect of capecitabine (CAP) treatment on the growth and metastasis of HCC. Methods Immunohistochemistry was used to detect PD ECGF protein levels in liver tissue of 61 patients with hepatocellular carcinoma (HCC). Northern blot was used to analyze the PD ECGF mRNA levels in 28 HCC tissues. Using 24 human nude mice high-metastatic models of liver cancer LCI D2 0, different doses of CAP were administered on the third day after tumor implantation. The nude mice were sacrificed on the third day after drug withdrawal. The length and length of the tumor were measured, and lung metastases were detected by HE staining. Results The positive rates of PD ECGF protein expression in 61 cases of HCC and pericancerous tissue were 70.5% and 47.5%, respectively. The positive rate of PD ECGF protein expression in HCC tissues of 37 cases with high TNM stage (stage III and IV) was 81.1. %) was significantly higher than the rate of expression of 24 low TNM staging groups (I and II) (5 4.2%), and the difference was significant (P < 0.05). NorthernBlot analysis showed that the expression level of PD-ECGF protein in HCC tissue was consistent with its corresponding mRNA level. The volume of liver cancer in the nude mice treated with CAP 2 .10mmol·kg-1·day-1 and 3.15mmol·kg-1·day-1 was significantly lower than that in the control group (24mm3± 119mm3 and 209mm3±15mm3). 15 9mm3) (P<0.05). Conclusion PD ECGF is highly expressed in HCC tissues and is associated with TNM staging of hepatocellular carcinoma. CAP inhibits the growth and metastasis of HCC.
