S期激酶相关蛋白2(S-phase k inase assoc iated prote in 2,Skp2)是泛素连接酶复合物的底物识别序列,对正常细胞周期调控起重要作用。在多数恶性肿瘤组织中Skp2呈过表达,并通过泛素化降解细胞周期蛋白依赖性激酶抑制剂p27k ip1,使细胞G1-S期进程加速。Skp2具有致癌潜能,与肿瘤的增殖,分化,抵抗凋亡和耐药性等生物学性状关系密切。Skp2不仅可作为判断肿瘤患者预后独立的指标,还可能成为肿瘤的药物和基因治疗的新靶点。
S-phase kase associated protein 2 (Skp2) is a substrate recognition sequence of ubiquitin ligase complex, which plays an important role in the regulation of normal cell cycle. Skp2 is overexpressed in most malignant tumors and the G1-S progression is accelerated by ubiquitination of the cyclin-dependent kinase inhibitor p27k ip1. Skp2 has carcinogenic potential and is closely related to biological characteristics such as proliferation, differentiation, resistance to apoptosis and drug resistance. Skp2 can not only be used as an independent index to judge the prognosis of cancer patients, but also be a new target of drug and gene therapy for tumors.
