目的:了解川芎内生放线菌群结构,筛选拮抗川芎根腐病原菌的放线菌资源。方法:采用组织块法、匀浆法、划线平板法等对川芎根茎中的放线菌进行分离和纯化,平板对峙法及抑菌圈法评价各菌株抑菌活性,显微观察法结合DNA测序进行菌种物种鉴别。结果:共获得83株川芎内生放线菌,分属13个类群;其中链霉菌属灰红紫类群中的深海链霉菌Streptomyces scopuliridis(KF600747.1)、灰锈赤链霉菌Streptomyces griseorubiginosus(AB706352.1)、团孢链霉菌Streptomyces agglomeratus(LC055413.1)能同时抑制4种川芎根腐病原菌。结论:内生放线菌群中的链霉菌属灰红紫类群是抑制川芎根腐病原菌的重要生防菌,在川芎药材生产中有着重要的应用前景。
Objective: To understand the structure of endophytic actinomycetes in Chuanxiong and screen for antagonistic actinomycetes resources of root rot pathogens. Methods: The actinomycetes in rhizome of Chuanxiong Houttuyniae were isolated and purified by tissue block method, homogenization method and scribing plate method. The antibacterial activity of each strain was evaluated by plate confrontation method and inhibition zone method. Microscopic observation combined with DNA Sequencing bacteria species identification. Results: A total of 83 strains of endogenous actinomycete were obtained, belonging to 13 groups. Streptomyces scopuliridis (Streptomyces scopuliridis) and Streptomyces griseorubiginosus (Streptomyces griseorubiginosus) from Streptomyces spp. 1), Streptomyces agglomeratus (LC055413.1) can simultaneously inhibit the four kinds of root rot pathogen Chuanxiong. Conclusion: Streptomyces reticulata in endophytic actinomycetes is an important biocontrol agent that inhibits root rot pathogen of Chuanxiong. It has an important application prospect in the production of Chuanxiong medicine.
