目的:分别测定6种葫芦科植物药材中总葫芦素、葫芦素B、葫芦素E的含量,比较不同药材中葫芦素类成分的含量差异。方法:建立UV-Vis分光光度法测定总葫芦素含量,显色剂:5%香草醛-冰醋酸-浓硫酸溶液,检测波长:543 nm。采用HPLC同时测定葫芦素B、葫芦素E的含量,流动相:乙腈-0.1%冰醋酸水溶液(49∶51),检测波长230 nm。结果:总葫芦素含量以木鳖子、栝楼最高,南瓜蒂最低;葫芦素B含量以甜瓜蒂最高,雪胆、栝楼、南瓜蒂均较低;葫芦素E含量以栝楼、甜瓜蒂最高,雪胆、南瓜蒂较低。结论:所建立UV-Vis分光光度法测定总葫芦素含量,简便、可靠、准确、重复性好。不同药材中葫芦素类成分含量有一定差异,可作为葫芦科植物药材质量评价、新药组方药材选择、中药药效机理研究的基本依据。
OBJECTIVE: To determine the contents of total cucurbitacin, cucurbitacin B and cucurbitacin E in six Cucurbitaceae herbs, and to compare the contents of cucurbitacins in different medicinal herbs. Methods: The total cucurbitacin content was determined by UV-Vis spectrophotometry. The color reagent: 5% vanillin-glacial acetic acid-concentrated sulfuric acid solution, detection wavelength: 543 nm. The contents of cucurbitacin B and cucurbitacin E were determined by HPLC. The mobile phase was acetonitrile-0.1% glacial acetic acid (49:51) and the detection wavelength was 230 nm. Results: The content of total cucurbitacin was the highest in Momordicae and the lowest in Fructus Punctatus, and the lowest in Cucurbita. The content of cucurbitacin B was the highest in Fructus Mume, but the content of Cucurbitacin E was lower in Fructus Schisandrae, Highest, snow, pumpkin low. Conclusion: The established total cucurbitacin content determined by UV-Vis spectrophotometry is simple, reliable, accurate and reproducible. The contents of cucurbitacins in different medicinal materials have certain differences, which can be used as the basic basis for the quality evaluation of cucurbits, the selection of new medicinal herbs and the pharmacodynamic mechanism of traditional Chinese medicine.
