目的 :探讨中药复方“脑梗通”对脑缺血后血浆和脑组织一氧化氮 (NO)含量的影响 ,了解药物的脑保护作用与NO含量变化的关系。方法 :用不同剂量脑梗通分别给Wistar大鼠灌胃 ,并与用生理盐水调制的活血通脉胶囊灌胃的同类大鼠做对照 ,连续 3d后建造右侧大脑中动脉阻断 (MCAO)局灶脑缺血模型并进行实验 ,观察血浆和脑组织中不同时相NO含量的变化。结果 :大鼠MCAO后 4h血浆及不同脑区的NO含量明显降低 (P <0 0 5) ,2 4hNO含量接近于正常组 (P >0 0 5)。脑梗通组在大鼠MCAO后上述 2个时相中血浆及脑组织NO含量有显著差异 (P <0 0 5) ,MCAO后 2 4hNO含量与对照组有显著差异 (P <0 0 5) ,但与正常组无显著差异 (P >0 0 5)。阳性对照组无上述差异性变化。结论 :在局灶脑缺血不同时相的NO含量不同 ,所起作用具有不同的病理生理意义 ;脑梗通可能通过促进NO合成并调控在正常范围而起保护神经细胞的作用。
Objective: To investigate the effect of Chinese herbal compound “Cerebral Infarction” on the content of nitric oxide (NO) in plasma and brain tissue after cerebral ischemia, and to understand the relationship between brain protective effect of drugs and the change of NO content. METHODS: Wistar rats were intragastrically administered with different doses of cerebral infarction, and compared with rats of the same type that were administrated with Huoxue Tongmai Capsule administrated with physiological saline. Right middle cerebral artery occlusion (MCAO) was established 3 days later. Focal cerebral ischemia models were performed and experiments were conducted to observe the changes of NO levels in plasma and brain tissues at different phases. RESULTS: The NO content in plasma and brain areas was significantly decreased (P < 0.05) at 4 h after MCAO in rats, and the NO level was similar to that in the normal group at 24 h (P > 0 05). There was a significant difference in NO content between plasma and brain in the cerebral infarction group after MCAO in rats (P < 0.05), and there was a significant difference in NO content between 24 hours after MCAO and the control group (P <0 05). However, there was no significant difference from the normal group (P > 0 05). The positive control group did not have the above-mentioned differences. Conclusion : The NO content in different phases of focal cerebral ischemia is different, which has different pathophysiological significance. Cerebral infarction may play a role in protecting nerve cells by promoting NO synthesis and regulating in the normal range.
