提出了一种快速、简便地测定Sn(Ⅱ ) -H2 O2 体系产生羟自由基 (·OH)的新方法。用荧光法建立了测定羟自由基的产生量 ,采用苯基荧光酮 phenylfluorone作为Sn(Ⅱ ) -H2 O2 反应体系的荧光试剂。通过测定苯基荧光酮反应前后的荧光强度变化 ,间接地测定羟自由基产生量。找到了体系最佳实验条件。利用电子自旋共振法ERS ,测定苯基荧光酮加入前后的波谱变化 ,从而进一步证明了该方法的可靠性。同时 ,测定了几种保健药物对羟自由基 (·OH)的清除能力。方法简便 ,测定快速 ,稳定性好 ,灵敏度高 ,无需昂贵的仪器 ,可作为一种筛选抗氧化剂的简便方法 ,亦可作为食品保鲜 ,化妆品性能检验的一种方法。
A new method for the determination of hydroxyl radical (· OH) in Sn (Ⅱ) -H 2 O 2 system is proposed. Fluorescence method was used to determine the amount of hydroxyl radical produced. Phenylfluorone phenylfluorone was used as the fluorescent reagent of Sn (Ⅱ) -H 2 O 2 reaction system. The amount of hydroxyl radical produced was determined indirectly by measuring the change in fluorescence intensity before and after phenylfluorone reaction. Found the best system conditions. The electron spin resonance method (ERS) was used to determine the spectral changes before and after addition of phenylfluorone, which further proves the reliability of the method. At the same time, the ability of scavenging hydroxyl radicals (· OH) by several health-care drugs was measured. The method has the advantages of simple method, rapid determination, good stability, high sensitivity and no expensive equipment, and can be used as a simple and convenient method for screening antioxidants, and can also be used as a method for food preservation and cosmetic performance testing.
