目的:观察清上蠲痛合剂对偏头痛大鼠的预防作用及对血清中β-内啡肽(EP)、降钙素相关基因肽(CGRP)、一氧化氮(NO)的影响。方法:分组方法及给药方式采用硝酸甘油(GTN)皮下注射法建立偏头痛大鼠模型,将成年健康SD大鼠36只,雌雄各半,体质量(188.29±7.60)g,随机分为正常组(生理盐水剂量:与等效剂量等容积),模型组(生理盐水剂量:与等效剂量等容积),清上蠲痛合剂预防组[等效剂量,7.46 g/(kg·d)],以上每组12只。预防组及模型组连续给药6 d后造模用药。除正常组外,其余各组采用皮下注射硝酸甘油方法复制大鼠偏头痛模型,观察其耳红、甩头或挠头、爬笼出现时间、次数及持续时间等行为学指标后,采用酶联免疫法吸附法(ELISA)测定各组大鼠血清中EP、CGRP、NO的含量。结果:与正常组相比,造模后模型组大鼠血清β-EP含量较正常组下降(P<0.001);清上蠲痛合剂预防组大鼠血清中β-EP含量较模型组明显升高(P<0.05)。造模后模型组大鼠血清CGRP和NO含量较正常组升高(P<0.001),清上蠲痛合剂预防组能抑制上述改变(P<0.05)。结论:清上蠲痛合剂能明显预防大鼠偏头痛发作,可能与调节机体CGRP、NO、EP的释放有关。

After modeling, the levels of serum CGRP and NO in model group were higher than those in normal group (P <0.001), and Qing Shang Tiao Tong mixture prevention group could inhibit these changes (P <0.05). Conclusion: Qing Shang Tiao Tong Mixture can obviously prevent the onset of migraine in rats, which may be related to the regulation of the release of CGRP, NO and EP.
