目的通过动物实验观察龙牡壮骨冲剂对小鼠胃肠动力及组织胃动素的影响,以探讨该药对胃肠动力的影响及机制。方法将小鼠随机分为对照组、模型组、龙牡壮骨冲剂大、中、小剂量组,用不同药剂复制胃肠动力紊乱模型,以营养性半固体糊灌胃,标记胃肠测量胃残留率和小肠推进比,同时以放免法测胃动素(MTL)水平。结果龙牡壮骨冲剂对正常组小鼠胃残留率和小肠推进比无影响,对胃复安所致胃排空加速、阿托品所致胃排空减慢、新斯的明所致小肠推进比增加、肾上腺素所致小肠推进比降低有逆转作用(均为P<0.05),与剂量有一定的相关性。结论龙牡壮骨颗粒可双向调节胃肠运动,并具有剂量依赖性,其对胃肠动力的调节作用可能是通过影响局部组织中的胃动素水平实现的。 Objective To observe the effects of Longmu Zhuanggu Granules on gastrointestinal motility and tissue motilin in mice by animal experiments to explore the effect and mechanism of the drug on gastrointestinal motility. Methods Mice were randomly divided into control group, model group, large, medium and small dose groups of Longmu Zhuanggu granules. The gastrointestinal motility disorder models were duplicated with different agents, and the gastrointestinal motility was administrated with nutritive semisolid paste. The residual rate and intestinal propulsion ratio were measured simultaneously with radioimmunoassay (MTL) levels. Results Longmu Zhuanggu granules had no effect on the gastric residual rate and the small intestine advancing ratio of mice in the normal group, but accelerated the gastric emptying caused by metoclopramide, slowed gastric emptying caused by atropine, and promoted the small intestine induced by neostigmine. Increased, epinephrine-induced reduction in the rate of intestinal propulsion was reversed (all P < 0.05), with a certain correlation with dose. Conclusion Longmu Zhuanggu Granules can regulate gastrointestinal motility in both directions, which is dose-dependent. Its regulation of gastrointestinal motility may be achieved by affecting motilin levels in local tissues.
