目的:建立离子对高效液相色谱法检测中成药及保健食品中添加二甲双胍、苯乙双胍、格列齐特、格列美脲、格列本脲、格列吡嗪、甲苯磺丁脲、罗格列酮、吡格列酮共9种化学降糖药。方法:采用AgilentZORBAX-EclipseXDB-18(4.6mm×250mm,5μm)色谱柱分离,流动相为0.02mol·L-1磷酸二氢钾溶液(含2mmol.L-1十二烷基硫酸钠)-乙腈(62.5∶37.5)(用1mol.L-1氢氧化钠溶液调节pH至6.5),流速1.0mL·min-1,检测波长230nm,柱温30℃。结果:9种化学降糖药能完全分离;具有宽的线性范围和良好的线性关系(r>0.9992),检出限为5.9~43.5μg.L-1。结论:该方法用于中药及保健食品中化学降糖药的检测具有快速、简便、高效的特点。
OBJECTIVE: To establish a HPLC-MS method for the determination of Chinese traditional medicine and health food supplemented with metformin, phenformin, gliclazide, glimepiride, glibenclamide, glipizide, tolbutamide, Glitazone, pioglitazone a total of 9 kinds of chemical hypoglycemic agents. METHODS: The mobile phase consisted of 0.02mol·L-1 potassium dihydrogen phosphate solution (containing 2mmol.L-1 sodium dodecyl sulfate) -acetonitrile (HPLC) with Agilent ZORBAX-EclipseXDB-18 (62.5: 37.5) (pH was adjusted to 6.5 with 1mol.L-1 sodium hydroxide solution). The flow rate was 1.0mL · min-1. The detection wavelength was 230nm and the column temperature was 30 ℃. Results: Nine kinds of chemical hypoglycemic agents could be completely separated, with wide linear range and good linearity (r> 0.9992). The detection limit was 5.9 ~ 43.5μg.L-1. Conclusion: The method is rapid, simple and efficient for the detection of chemical hypoglycemic agents in traditional Chinese medicine and health food.
