以耐寒性不同的鲁果12号和香玲核桃品种为试材,在-15、-20、-25、-30、-35℃低温处理下,研究了离体枝条的质膜相对透性、膜脂过氧化程度、过氧化氢(H2O2)含量以及抗坏血酸-谷胱甘肽(AsA-GSH)循环中的相关物质含量及酶活性等指标,分析其耐寒机理。结果表明,低温胁迫明显增加了核桃枝条的膜透性、超氧阴离子自由基(O2-·)产生速率、丙二醛(MDA)和H2O2含量,但耐寒品种鲁果12号的增幅低于香玲;低温胁迫期间,鲁果12号枝条中抗坏血酸(AsA)、谷胱甘肽(GSH)含量以及AsA/DHA值和GSH/GSSG值的降幅低于同温度处理的香玲,脱氢抗坏血酸(DHA)、氧化型谷胱甘肽(GSSG)含量的增幅低于同温度处理的香玲;抗坏血酸过氧化物酶(APX)、单脱氢抗坏血酸还原酶(MDHAR)、脱氢抗坏血酸还原酶(DHAR)、谷胱甘肽还原酶(GR)活性的增幅高于同温度处理的香玲。表明鲁果12号在低温胁迫期间能够保持较高的AsA-GSH循环效率,可有效抑制H2O2的积累,降低活性氧的伤害。
The relative plasma membrane permeability of detached shoots was studied at low temperature of -15, -20, -25, -30 and -35 ℃ with different cold resistance of Luogu 12 and Hongling walnut cultivars Lipid peroxidation, hydrogen peroxide (H2O2) content and related substances in ascorbate-glutathione (AsA-GSH) cycle and enzyme activity were analyzed. The results showed that under low temperature stress, the membrane permeability, the production rate of O2- ·, the contents of malondialdehyde (MDA) and H2O2 in walnut shoots were significantly increased, but the growth of hardy variety Luogu 12 was lower than that of Ling; At low temperature stress, the decrease of AsA and GSH and the decrease of AsA / DHA and GSH / GSSG in Luogu 12 were lower than those of Linglin and dehydroascorbic acid DHA and GSSG were lower than that of the same temperature treatment. Ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) ), Glutathione reductase (GR) activity increased more than the same temperature treatment of Hong Ling. The results showed that Luguo 12 can maintain a high AsA-GSH cycle efficiency during low temperature stress, which can effectively inhibit the accumulation of H2O2 and reduce the damage of reactive oxygen species.
