用超声波法提取鸡油菌多糖CCP,经浓硫酸法对其进行硫酸化修饰,得到硫酸酯鸡油菌多糖S-CCP。通过测定对超氧阴离子、羟自由基的清除率,以及对H2O2诱发MDA生成的抑制和H2O2诱发红细胞溶血的影响,评价S-CCP的体外抗氧化作用。实验结果表明,S-CCP较之CCP具有明显的体外抗氧化作用。当多糖浓度为5mg/mL时,S-CCP对O2·清除率高出CCP22.71%;多糖浓度0.4mg/mL时,S-CCP对·OH清除率高出CCP14.9%;多糖浓度4mg/mL时,S-CCP作用于H2O2诱发MDA的生成量低于CCP0.4nmol/mg,且其对H2O2诱发红细胞氧化溶血抑制率高出CCP33.9%。
Ultrasound extraction of chanterelles polysaccharide CCP, sulfuric acid by sulfuric acid method to be modified to give chanterelles polysaccharide S-CCP. The anti-oxidative effects of S-CCP in vitro were evaluated by measuring the clearance rate of superoxide anion and hydroxyl radical, inhibiting H2O2-induced MDA production and H2O2-induced erythrocyte hemolysis. The experimental results show that S-CCP has obvious anti-oxidative effect in vitro compared with CCP. When the concentration of polysaccharides was 5mg / mL, the scavenging rate of O2 · was 22.71% higher than that of S-CCP; the scavenging rate of · -OH by S-CCP was 14.9% higher than that of polysaccharides when the concentration of polysaccharides was 0.4mg / mL; / mL, the production of S-CCP by H2O2-induced MDA was lower than CCP0.4nmol / mg, and the inhibitory rate of H2O2-induced erythrocyte oxidation and hemolysis was 33.9% higher than that of CCP.
