本文通过实验,并结合Gen Bank数据库序列,分析ITS、ITS2、psb A-trn H、mat K、rbc L 5条常用条形码共978条序列在种内与种间变异、barcoding gap、系统发育树等方面对百合属植物的鉴别能力,并采用最近距离法(nearest distance)和相似性搜索算法(BLAST1)评价5条序列的鉴定成功率。研究结果显示,不同条形码序列可以较好的鉴别百合属内不同物种。ITS序列鉴定成功率较高,种内种间差异显著,系统发育树可以很好地将不同物种分开。结果表明DNA条形码技术可以准确地鉴别百合属植物,ITS序列可以作为鉴别百合属不同物种的优选序列。
In this paper, 978 sequences of 5 common bar codes of ITS, ITS2, psb A-trn H, mat K, and rbc L were analyzed through experiments and GenBank database sequences in intraspecies and interspecies variation, barcoding gap, phylogenetic tree, etc. The identification ability of Lilium was evaluated. The success rate of identification of five sequences was evaluated by using the nearest distance and similarity search algorithm (BLAST1). The results show that different barcode sequences can better identify different species in Lilium. The success rate of ITS sequence identification is high, and there is significant difference among species within species. Phylogenetic tree can separate different species well. The results showed that DNA barcode technology can accurately identify Lilium, ITS sequence can be used as a preferred sequence to identify different species of Lilium.
