目的 :比较脂质体rIL - 2和游离rIL - 2分别联合肿瘤浸润淋巴细胞治疗鼠H2 2肝癌的效果及副作用。方法 :采用薄膜分散法和超声波粉碎处理法制备脂质体rIL - 2 ;选昆明鼠 40只 ,随机分组进行体内治疗 ,游离rIL - 2用量为脂质体rIL - 2的 3倍 ,观察瘤出现时间、瘤体积、瘤湿重及鼠体温的变化。结果 :TIL +rIL - 2组的瘤出现时间、瘤体积、瘤湿重分别为 :15± 2 .5d、2 .44± 2 .97cm3 、4.7± 2 .0 g ;TIL +脂质体rIL - 2组分别为 :15± 3.2d、2 .2 7± 2 .5 8cm3 、4.5± 2 .1g ;二者与对照组比较均显出了肯定的治疗效果 (P <0 .0 5 ) ,但二者之间比较无统计学差别 (P >0 .0 5 )。TIL +rIL - 2组有明显发热反应 ,TIL +脂质体rIL - 2组体温正常。结论 :脂质体rIL - 2较游离rIL - 2抗肿瘤作用提高 ,副作用降低。
Objective: To compare the efficacy and side effects of liposome rIL - 2 and free rIL - 2 in combination with tumor infiltrating lymphocytes in the treatment of murine H 2 2 hepatocellular carcinoma. Methods: Liposomes rIL - 2 were prepared by membrane dispersion method and ultrasonic pulverization method. Forty Kunming mice were selected and randomly divided into groups for in vivo treatment. The amount of free rIL - 2 was 3 times of that of rIL - 2. Time, tumor volume, tumor wet weight and body temperature changes. Results: The tumor appearance time, tumor volume and tumor wet weight of TIL + rIL - 2 group were 15 ± 2.5 days, 2.44 ± 2 .97 cm3 and 4.7 ± 2.0 g respectively. TIL + liposome rIL - The two groups were 15 ± 3.2d, 2.27 ± 2 .5 8cm3 and 4.5 ± 2.1g, respectively. Both of the two groups showed positive therapeutic effect (P <0.05) There was no significant difference between the two (P> 0.05). TIL + rIL - 2 group had a significant fever response, TIL + liposomal rIL - 2 body temperature was normal. Conclusion: The antitumor effect of rIL - 2 liposome is better than that of free rIL - 2, and its side effects are decreased.
