目的:研究丹红注射液(DHI)对斑马鱼节间血管(ISV)的促进作用,测适宜的药效指标值,为DHI谱-效相关质量评价系统的建立提供药效学基础。方法:通过PTK787抑制血管内皮生长因子表达的方法,建立斑马鱼ISV损伤模型,通过模型组中不同浓度DHI的加入,观察斑马鱼ISV生长状态,确定加药浓度。按照预实验的造模和给药方法,检测各给药组ISV长度、计算ISV生成率,考察DHI对促进斑马鱼ISV生长的作用。结果:PTK787组ISV长度均下降,与正常对照组相比有显著性差异(P<0.05)。加DHI治疗各组ISV长度增长明显,与PTK787组比较有显著性差异(P<0.05),30μL DHI组与正常对照组相比无统计学差异。不同批次DHI促血管生成率在40%-80%之间。结论:DHI对斑马鱼ISV具有明显的再生功能,对血管损伤有修复保护作用,方法可靠、数据可信,结果稳定。
Objective: To study the promotion effect of Danhong injection (DHI) on zebrafish interventricular blood vessels (ISV) and to find suitable pharmacodynamic indexes to provide a pharmacodynamic basis for the establishment of DHI spectrum-related quality evaluation system. Methods: The model of ISV injury was established by inhibiting the expression of vascular endothelial growth factor (PTM) by PTK787. Observing the growth status of zebrafish ISV by adding different concentrations of DHI in the model group, the drug concentration was determined. According to the pre-experimental modeling and administration method, the length of ISV in each administration group was measured, the formation rate of ISV was calculated, and the effect of DHI on the growth of zebrafish ISV was investigated. Results: The length of ISV decreased in PTK787 group, which was significantly different from that in normal control group (P <0.05). Compared with PTK787 group, the ISV length increased obviously in DHI group (P <0.05), but there was no significant difference between 30μL DHI group and normal control group. Different batches of DHI pro-angiogenesis rates between 40% -80%. Conclusion: DHI has obvious regeneration function on zebrafish ISV, which can repair and protect vascular injury. The method is reliable, the data is credible and the result is stable.
