用平衡透析法研究了生理条件 (pH 值7.43) 下Zn (Ⅱ) 与人血清白蛋白 (HSA) 或牛血清白蛋白(BSA) 的相互作用。结果表明, HSA对Zn (Ⅱ) 有1.1 个强结合部位和9.0 个弱结合部位, 结合常数分别为1.04×105 及1.67×103 ; BSA 对Zn (Ⅱ) 有1.2个强结合部位和约9.7 个弱结合部位, 结合常数分别为8.2×104 及1.9×103 。Zn(Ⅱ)-HSA和Zn(Ⅱ)-BSA的Hill系数分别为0.87、0.84,表明Zn(Ⅱ)与HSA或BSA的结合均只产生较小的负协同效应。
The interaction between Zn (Ⅱ) and human serum albumin (HSA) or bovine serum albumin (BSA) under physiological conditions (pH 7.43) was studied by equilibrium dialysis. The results showed that HSA had 1.1 strongly binding sites and 9.0 weakly binding sites on Zn (Ⅱ) with binding constants of 1.04 × 105 and 1.67 × 103, respectively, and BSA had 1 .2 strong binding sites and about 9.7 weak binding sites with binding constants of 8.2 × 104 and 1.9 × 103, respectively. The Hill coefficients of Zn (Ⅱ) -HSA and Zn (Ⅱ) -BSA were 0.87 and 0.84, respectively, indicating that the combination of Zn (Ⅱ) and HSA or BSA produced only a small negative synergistic effect.
